In vivo regeneration of rat laryngeal cartilage with mesenchymal stem cells derived from human induced pluripotent stem cells via neural crest cells

چکیده

The laryngotracheal cartilage is a cardinal framework for the maintenance of airway breathing, which occasionally requires reconstruction. Because hyaline has poor intrinsic regenerative ability, various approaches have been attempted to regenerate cartilage. use autologous mesenchymal stem cells (MSCs) regeneration widely investigated. However, long-term culture may limit proliferative capacity. Human-induced pluripotent cell-derived MSCs (iMSCs) can circumvent this problem due their unlimited This study aimed investigate efficacy iMSCs in thyroid immunodeficient rats. Herein, we induced through neural crest cell intermediates. For relevance prospective future clinical application, induction was conducted under xeno-free/serum-free conditions. Then, clumps fabricated from an iMSC/extracellular matrix complex (C-iMSC) were transplanted into defects Histological examinations revealed cartilage-like regenerated tissue and human nuclear antigen (HNA)-positive surviving lesion. HNA-positive co-expressed SOX9, type II collagen identified around cells. These results indicated that C-iMSCs promoted some differentiated chondrogenic lineage Induced be promising candidate therapy

Isolation and Characterization of Human Induced Pluripotent Stem Cells-Derived Mesenchymal Progenitors

Purpose: Isolating human induced pluripotent stem cells (hiPS)-derived mesenchymal progenitors as a new source of mesenchymal cells which can differentiate into different lineages like adipose and bone. Materials and Methods: After 7 days of hiPS1 culture on matrigle coated dishes, spindle like cells around colonies were removed by cell scraper. These cells that had mesenchymal like morphology ...

Gene Expression Profile of Adherent Cells Derived From Human Peripheral Blood: Evidence of Mesenchymal Stem Cells

Mesenchymal stem cells (MSCs) provide a novel option in cellular therapy and tissue engineering. Recent studies indicated that it is possible to obtain MSCs from peripheral blood by attachment ability to plastic surface. To evaluate adherent cells derived from peripheral blood, their expression profile and surface markers were investigated. The results of RT-PCR indicated that these cells expre...

Alpha-Tocopherol increases the proliferation of induced pluripotent stem cell derived neural progenitor cells

In addition to its antioxidant effect, Vitamin E or α–tocopherol is suggested to enhance remyelination in the animal model of non-inflammatory demyelination. In this study, the possible proliferative effect of vitamin E on human- induced pluripotent stem cell-derived neural progenitors (hiPS-NPs) and the underlying mechanisms were investigated in vitro. NPs were induced from iPS cells via 3 ste...

Induced Pluripotent Stem Cells: Challenges and Opportunities

Regenerative capacity of mammals is limited and can rarely regenerate a specific organ or tissue fully. Due to these limitations, regenerative medicine seeks efficient and safe cell sources for regeneration of damaged tissues and organs or treatment for incurable diseases. Human embryonic stem cells (HESCs) hold two important properties called self renewal and pluripotency. However, the use of ...

Functional mesenchymal stem cells derived from human induced pluripotent stem cells attenuate limb ischemia in mice.

BACKGROUND Aging and aging-related disorders impair the survival and differentiation potential of bone marrow mesenchymal stem cells (MSCs) and limit their therapeutic efficacy. Induced pluripotent stem cells (iPSCs) may provide an alternative source of functional MSCs for tissue repair. This study aimed to generate and characterize human iPSC-derived MSCs and to investigate their biological fu...
